You can either do it cold turkey or gradually move the 2am pumping session closer to your next pumping session.
I would probably start now and move it by an hour for a week at a time. That would help with the discomfort.
As for supply, it sounds like you have a good supply established so you should be fine. If you are finding you are not getting enough the you can always add that session back in.
